MES Hydrate (2-(N-morpholino)ethanesulfonic acid monohydrate) is one of the most widely used and well-characterized biological buffers in plant tissue culture, appearing as a standard component across decades of foundational literature and classical media formulations. As a member of the Good’s buffer family — a group of buffers specifically developed for biological research — MES offers several advantages that make it particularly well-suited to plant cell and tissue culture applications. Unlike inorganic buffers such as phosphate, MES does not act as a nutrient, interfere with metal ion availability or precipitate with common media components, making it especially valuable in formulations where micronutrient availability and pH stability are both critical. It is also membrane-impermeant and biologically inert at working concentrations, meaning it exerts minimal effect on cellular physiology — an important consideration when working with sensitive explants or regeneration protocols. MES Hydrate has an effective buffering range of approximately pH 5.5 to 6.7, with a pKa of around 6.1 at 25°C. This range maps almost perfectly onto the slightly acidic pH conditions favored by most plant tissue culture media, making it the go-to choice when reliable pH control throughout culture is required.
